研究方向研究室特色

蛋白质体学研究是指全面性的探讨蛋白质表现程度的学门。在生物医学领域中,通常着重於找出两个样品之间相对差异表现的蛋白。由於分析技术的快速进步,使得蛋白质体分析的覆盖率,已逐渐赶上基因体学研究。其中最主要的两项技术為质谱学及胜肽分离技术。本实验室主要的研究兴趣就是针对这两项技术进行研发及改进,以提升蛋白质体数据的广泛度、灵敏度、可信度及自动化程度。
对於肽分离系统的改进,我们建立了多种奈米级多维液相分离系统,不论是一般性的整体蛋白体分析,或是特定族群胜肽的纯化分离,其效能均已达国际一流的水準。利用这些系统,我们可以从30ug的细胞蛋白萃取物中鑑定出6000-9000个蛋白,甚至可从200微克蛋白中鑑定出近万条磷酸化肽。至於在质谱测量方面,我们着重於开发崭新的同位素标定技术,以提高质谱定量数据的可靠性。目前我们已建立两种标定技术,其中之可进行整体蛋白体定量分析,另一则专注的定量特定胺基酸的氧化程度。这些技术都将运用於疾病蛋白体学研究,尤其将着重於分析临床的组织或体液样品。
