研究方向:
(1)以整合性体学研究模式研发口腔癌早期诊断生物标记及精準治疗标靶 口腔癌為国人好发癌症之一,男性发生率世界第一,已成為国人重大健康议题与医疗负担。然而在过去二十年间,口腔癌的早期侦测与精準标靶治疗进展非常有限,目前仍无有效的生物标记(biomarker)可应用於提高早期口腔癌检出率及辅助术后追踪,也缺乏针对口腔癌研发的标靶治疗(target therapy)策略以提升个人化的治疗效果。本实验室带领分子医学研究中心研究团队,参与美国领导的(国际癌症登月计画的一重要部分),利用已建置成熟的多体学(multi-omics)分析技术平台,系统性深入研究国人口腔癌(病患癌组织与癌旁非肿瘤组织)的基因体(全外显子体及RNA定序)、蛋白体(定量蛋白体及磷酸化蛋白体)与代谢体(定量代谢体)全貌及其相关变异,透过整合性体学大数据分析以瞭解基因体变异所造成的蛋白体/代谢体变化,希望建构首个口腔癌 “多体学蓝图”(基因体→蛋白体→代谢体),提供探索这些系统性变异对口腔癌发生进展的生物意义;同时结合标靶基因/蛋白质/代谢物定量技术及人源化肿瘤动物模式,研发可应用於作為早期侦测之生物标记与精準医疗用药标靶或指引。
(2)利用先進蛋白质體學分析技術研发可临床应用之癌症生物标记 癌症為人类主要死因之一。大部份癌症被诊断时皆為晚期,是癌症死亡率居高不下的主因。除了养成健康生活习惯外,早期诊断与有效治疗是目前对抗癌症的唯一有效方法。虽然有效的癌症生物标记(biomarker)並不多,抽取体液(血液、尿液、唾液等)檢查其中可能的癌症生物標記仍是最方便的初步篩檢方法。因此,找尋新穎且更為有效的体液生物標記是對早期診斷癌症非常重要的課題。以質譜學為基礎之蛋白质體學為近年來新興學門,由於具備高靈敏度、高精確度與高通量之特性,被學界寄予厚望可以用來系統性分析(包括定性与定量)癌症相關樣本中蛋白质,藉以找尋與驗證具高應用價值的癌症生物標記。本實驗室與本校多位同仁合作,共同建立,发展多種蛋白质體相關定性與定量分析技術平台,分別利用三種生物材料(癌症病患、及)找寻癌症生物标记及分析癌症之异常分子调节机转。本实验室目前针对国人数种好发癌症,包括口腔癌、大肠癌、肝癌及胰臟癌同步进行研究,長期目標在於發展出可實際應用於臨床偵測早期癌症及其復發的体液生物標記,同時發展可快速方便檢驗這些生物標記的方法。目前已於2016/10发表,领先其他国际团队,已获多国专利,并技术移转给本土新创生技公司,进行后续免疫检测试剂开发(ELISA及试纸型快筛)与临床试验,此产学合作成果亦获得生技相关奖项的肯定,包括 「」(2019/07)、「」(2019/11)、 「」(2021/08) 及合作新创生技公司获「第16屆国家新創獎-初创公司奖」(2019/12)。该ELISA试剂组已於2020/12/04完成欧盟註册 (註册号00159706),产物已可以进入欧盟销售。此為国人自行成功开发的第一个口腔癌生物标记免疫检测商品化试剂组,可為后续一系列衍生或相关检测产物重要参考品,具指标性意义,对提升防治国人口腔癌的检测能力预期将有重大影响。而也已於2011/05发表,亦获多国专利及技术移转给本土新创生技公司,进行后续免疫检测试剂开发与临床试验。
(3)研發重要醫用或研究用蛋白质的核酸适体(Aptamer) 核酸适体(Aptamer)是具有独特立体结构的单链RNA或DNA(长度在20~80个硷基)、可与特定分子标靶以构型互补模式结合,此结合作用与抗体--抗原反應類似,因此核酸适体具有發展成為與抗體類似的拮抗劑或分子靶向工具的高應用潛能。本實驗室已建立以PCR為基礎的核酸适体的高效篩選技術SELEX(systematic evolution of ligands by exponential enrichment),將針對重要醫用或研究用蛋白质開發高親和力核酸适体,提供研发疾病检测或标靶治疗的新颖分子工具。 ? (4)應用蛋白质體學技術研究細胞訊息傳遞分子機制 本實驗室以累積多年之蛋白质酵素分離純化鑑定經驗,過去十多年中投入並成功研發以質譜學為基礎之多種蛋白质體相關定性與定量分析技術。利用這些強大效能蛋白质體分析工具,我們可以重新擴大研究檢視各種細胞內關鍵訊息傳導路徑、發現新穎蛋白质交互作用及分析各種蛋白质後轉譯修飾作用,這些可能性大幅提升了細胞生物學領域研究的廣度與深度。 ? |
