高通量(high-throughput)生物实验技术揭开后基因体世代 (post-genomic era)的序幕,而近年呈现爆炸性发展的次世代定序生物技术,更进一步将生物实验技术推向高效率、高精度、高準确性之境界,為生命科学研究领域创造极佳的契机。次世代定序方法目前也已被广泛应用於各类型的生命科学研究工作上,研究人员可针对DNARNA进行高速定序,搭配后端生物资讯 (bioinformatics)的应用,进行基因序列变异分析、基因结构异常分析、基因表现量分析、微小RNA (small RNA)表现量分析、基因体甲基化修饰等之研究。

本实验室的研究主要在建立和应用各种NGS技术平台於癌症基因体及转录体上的研究搭配生物资讯的分析, 系统性的找出癌症重要的“生物特徵”相关病理特徵,利用大數據庫運算,建構其可能參与之調控網絡,并利用细胞和动物模式进行深入的分子机制探讨及验証。近年主要研究包括:

  1. 建立台湾本土癌症组织之功能性基因體體學數據庫

建立完整的台湾本土癌症组织(口腔癌/食道癌/大肠直肠癌/脑癌)之功能性基因体体学数据库,并利用生物资讯方式,结合国外大数据库系統性的分析找出与臨床的早期診斷、疾病的治療与用藥、抗藥性、存活率与再復性相关的重要分子,完整的描繪這些變體的病理作用機制以及調控網絡。針對其中重要的變異基因,進一步使用細胞和動物模式研究來深入探討及驗証這些癌症相关標的之分子機制。

  1. 开发分析非编码RNA调控网络之生物资讯工具

利用非编码RNA已知之生物特性 (long noncoding RNA/circRNA/miRNA),结合大数据库,开发分析非编码RNA调控网路之生物资讯分析平台。进一步结合细胞和动物模式,验証其中新颖之非编码RNA的调控分子机制。

  1. 开发次世代定序法於癌症液态检体之应用

外泌體組成不僅反應了起源細胞,且富含与細胞轉化相关的生物分子,因此已成為近年來液體活檢方法中最有潛力的目標。實驗室建構了一套只需1 cc的血浆,即可快速提取外泌核糖核酸方法,利用次世代定序法验証了此方法明显优化并提升样本製备的稳定性。我们更进一步全面性的建立了健康人及多种癌症之外泌微核糖核酸体学数据库 (口腔癌、食道癌、肺癌、大肠直肠癌),结合临床资讯,系统性的寻找具早检测、长期监测治疗反应以及预后之多面向的生物标誌。因应精準医学方面之应用,更进一步开发一套快速检测晶片系统,可精準且多面向的监测癌症及治疗进程。利用多种生物资讯工具,完整描绘出外泌微核糖核酸在癌症进程所扮演的功能角色,并在细胞及动物模式上进行深入的机制研究。
