人类如何将学习的经验转為记忆储存一直都是科学界最想解开的谜题。人类拥有高度学习与记忆的能力,使人类祖先能在恶劣的自然环境中生存,进而成為优势物种。俗话说:”一朝被蛇咬,十年怕草绳就是一个例子。先前被毒蛇咬过而大难不死的人,将毒蛇的样子与危险讯号两种讯息关联在一起并转变為记忆储存,下次再看到相似外型的物体,先前储存的记忆便会被唤起,出现恐惧的感觉进而产生警戒。而这些记忆的形成是由大脑上千亿颗神经元共同运作的结果,但确切机制目前仍然未知。我们研究室利用果蝇作為模式生物了解大脑在学习与记忆过程中产生哪些生理变化。果蝇大脑仅有约十叁万颗神经细胞,相较於人脑由将近千亿颗神经细胞组成相对简单。我们利用大脑组织免疫染色搭配共軛焦显微镜技术观察果蝇大脑内神经分子的变化情形,进而了解记忆过程中大脑的分子变化情形。搭配活体大脑钙离子影像系统研究果蝇大脑在学习前与学习后的神经反应变化情形。另外利用动物行為学分析观察特定基因突变或特定神经细胞的神经活性改变是否影响果蝇学习与记忆能力。这些实验结果都将使我们更加了解记忆形成的大脑神经机制。
