研究方向及研究室特色
我们实验室之研究方向為开发侦测及定量物生物分子(包括蛋白质及代谢物)之质谱技术,这些方法已经被用来研究细胞模式及临床组织及体液等真实样品,并经由国际合作保持技术的新颖性。我们的研究目标在於利用本实验室之专长(蛋白质体、代谢体及生物质谱技术)了解泌尿系统癌症之生物机制,利用体学的方法研究膀胱癌及肾臟癌细胞株、肿瘤组织及尿液,希望找寻早期诊断癌症之非侵入式生物标记。
并与泌尿科、肾臟科、妇产科、小儿科等临床医生合作,运用蛋白质体及代谢体等后基因体学研究方法,全面性的研究正常与疾病生物系统(细胞株及临床收集的体液或组织)中蛋白质与代谢物之群体变化,监测各个分子在疾病所扮演的角色,探讨恶性与良性泌尿系统疾病发生及恶化之生物机制,以期朝向临床应用及个人化医疗迈进。
目前实验室的研究方向如下 :
1. 以系统性方法找寻及验证泌尿系统癌症之生物标誌及其生物重要性

我们研究了膀胱癌细胞株之分泌蛋白质体以及临床医生取得之肿瘤及尿液组成蛋白体,经由交叉比对,已经发现一系列可能随着癌症的发生而产生表现量改变的分子,未来将持续验证其重要性,并研究其与病理状况之相关性,了解其在泌尿系统研症所扮演之角色。

2. 发展标靶性及多重定量特性之MRM-MS技术在大量临床样品中验证疾病生物标誌之重要性 (Targeted and Multiplexed Quantitation of Proteins for Verification of Biomarker Panels using Multiple-Reaction-Monitoring MS (MRM-MS) and immune MRM-MS. 蛋白质体学是当今疾病生物标誌研究中最热门的研究领域之一。我们研究团队过去的研究成功地以iTRAQ技术分析发现膀胱癌病患尿液检体中多种蛋白质生物标誌,同时也已建立「液相层析多重反应监测质谱定量分析技术」(LC-MRM-MS),首度完成於人类临床尿液样本中同步定量63个蛋白质分子的研究工作,此技术将可协助研究学者在大量检体中验证过去所找到的生物标记后选分子,未来将继续发展「免疫浓缩-液相层析多重反应监测质谱定量分析技术」(immuno-MRM-MS)及生物资讯分析方法,以扩展至其他疾病或不同型态的检体。

随着标靶定量技术的发展,我们开始可以利用质谱方法大规格的追踪参与特定生物反应机制之多种蛋白质及代谢分子,比较细胞模式在不同处理条件下的表现量,监测各个分子在疾病所扮演的角色,并在个人临床样品中验证,以期朝向个人化医疗迈进。论文与着作
