本实验室以过敏相关疾病為研究主轴,研究疾病的分子机制及开发新疗法。过敏与气喘疾在已开发国家盛行率愈来愈高,病严重影响人民的健康与消耗许多医疗资源,这类疾病是由多个遗传与环境因子引起体内免疫系统不平衡反应所致,我们的研究重心主要在於深入探讨可以调控过敏病的分子与细胞间反应,期许应用於研发新的防治方式。

气喘病之基因治疗:基於气喘常為第二型辅助细胞过度反应所致,嚐试利用病毒载体携带抗发炎分子或第一型辅助细胞相关之细胞激素基因,经由气道施予疾病模式小鼠,用以平衡过度活化的第二型辅助细胞免疫反应;或是携带可干扰第二型辅助细胞反应的基因片段,评估其抑制气喘疾病各项指标的可行性。

药物或是抗体引发調節細胞的發展:近年,如何調節過度活化的免疫反應成為免疫学的熱門課題,我們亦著手研究與表觀遺傳學的基因表達調控相關藥物、或是可改變辅助细胞族群活化功能的特殊抗体等,对人类周边血球中的被激活辅助细胞或是气喘疾病模式小鼠的免疫调节功能。在逐步研究这些药物如何调控气喘疾病发生的分子机制过程中,可以进一步发展运用这些药物或抗体来治疗气喘疾病。

探讨可治疗气喘中药之科学证据:以临床使用疗效佳之方剂如「定喘汤」,利用气喘动物模式来研究中药使用之剂量、给药时机、疗效机制及安全性,做為分离出有效成分之基础;也和中、西医临床医师合作,完成疗效评估。在中医药科学化方面,则将注重中药作用於细胞讯号传递系统之机制,并搭配以中草药材中的纯化药物,来探讨并分析各药物对周边淋巴球细胞活性的调控或是对气喘疾病模式小鼠的抑制发炎功能。

脐带血淋巴细胞活性研究:比起骨髓,移植脐带血中造血系统,有材料取得的方便性与排斥性低的优点。所以本实验室的另外一个研究主题,是和长庚儿童医院医师合作,探讨以不同细胞激素组合对脐带血中 T 辅助细胞及自然杀手细胞之复製、死亡及活性的调控,或是评估不同免疫抑制剂对细胞功能的影响,可以对脐带血移植时的免疫反应,有更多的了解。
