壹、肺癌标誌蛋白质的开发与功能性探讨

肺癌是台湾也是全世界最主要的癌症死亡原因,其中非小细胞肺癌是属於高发生率与高致死率的癌症。我们实验室致力於发展有助於非小细胞肺癌诊断/预后与治疗的肿瘤标誌(tumor marker) 蛋白质。此外,我们也进一步探讨关键肿瘤标誌蛋白质的分子作用机转,期待能為肺癌找寻新的治疗方向。我们运用的材料包含肺癌组织,体液 (血清,肋膜积液以及肺癌细胞株。主题包括: ()建立与肿瘤分期相关的组织蛋白体资料库以寻找与转移相关的蛋白质,用以评估肺癌病人的预后与治疗 ()发展体液中,有助於非小细胞肺癌诊断/预后的肿瘤标 ()深入探讨肺腺癌标誌蛋白质的分子作用机转,其中专注於 KPNA2 这个具有发展潜力的致癌分子的一系列研究,包含 (a). 研究 KPNA2 蛋白质在肺腺癌细胞中的讯息联络网 (b). 分析与肿瘤侵犯力有关的 KPNA2蛋白质复合体成员及其作用机转 (c). 找寻调节KPNA2 在癌症细胞过度表现的讯息传递路径与转录因子,以协助肺癌标靶治疗的开发

贰、探讨人类 Arl1 蛋白质复合体及其生理功能

腺嘌呤核苷二磷酸核糖化相似因子 (Arf-likeArl)  Ras small GTPase 超级家族的成员。Arl1 主要位於反式高基氏体网路上,藉由调节其作用分子 (effector)参与细胞内的囊泡运输 (vesicle transport)。然,关於 Arl1 及其 effector 的详细分子作用机转与扮演的生理功能则尚未釐清。我们利用蛋白质体学方法系统性的鑑定 Arl1 蛋白质复合体成员并验证 Arl1 与其作用分子 (effector) 共同参与囊泡运输的分子机转并探讨 Arl1 及其作用分子生理意义。
