研究室特色



分子调控暨生物资讯研究室之主要研究目标為以单细胞生物阴道鞭毛虫作实验材料,探讨细胞寿命延长及化的调控机转。研究特色是利用生物资讯整合各种经由高通量分析平臺所产生之数据对基因体、转译体,蛋白质体作為基础,并从此等资料中进行探磡,发掘可能调命单细胞原生动物寿命延长及化的机转。

研究方向:阴道鞭毛虫微小蛋白调控缺铁反应之研究

微小蛋白质是由少於一百个胺基酸所组成的蛋白质,最近的研究显示这群蛋白质参与许多细胞内反应,如肿瘤生成、压力反应及讯息传递等。然而,因為传统的基因註解方式将一百个胺基酸的长度作為一个阀值,将转译出短於一百个胺基酸的开放阅读框排除,导致微小蛋白一直是被忽略的一群。

基於近年发展成熟的次世代定序技术,其应用广泛,不仅让微小蛋白质得已被鑑定出来,更藉此能获得更全面的基因表现资讯。然而截至目前為止,微小蛋白质尚未在任何原虫中被发现。本研究室将利用阴道鞭毛虫作為一个模式生物,鑑定并探讨微小蛋白质在缺铁压力下所扮演的角色。阴道鞭毛虫是造成人类非病毒性、盛行率最高的性接触传染病阴道鞭毛虫症的病原体。铁离子对於调控阴道鞭毛虫细胞内能量代谢、氧化还原平衡及氢化酶体质量等相当重要,因此阴道鞭毛虫如何在缺铁情况下生存是相当重要的课题。经由全基体开放阅读框、第二代/第叁代高通量定序及蛋白质体学整合性生物资讯分析,已初步确认在阴道鞭毛虫的转录体及蛋白体中可发现新的微小蛋白。这初步研究结果开启了一个全新的研域。因此未来计画的目标為研究阴道鞭毛虫的微小蛋白质在缺铁情况的功能性角色,并且建立一套针对微小蛋白质的实验及分析平台。
