研究方向及研究室特色

  1. 在细胞中,基因表达仰赖严谨的转录调控。此过程由 Mediator 复合体、RNA 聚合酶 II 及多种转录相关复合体等巨分子机器共同协调,确保转录能够精準执行并维持细胞正常运作。一旦这些复合体出现结构或功能异常,便可能导致癌症与其他疾病。然而,这些生物大分子不仅结构高度复杂,亦展现多样的构型,其精细的调控机制至今仍有许多未解之处。深入理解其分子运作原理,对於疾病研究与治疗策略的发展至关重要。

本实验室专注於真核基因转录调控研究,特别聚焦於 Mediator 复合体及其激酶模组的结构与功能,并探讨其与多种转录因子及细胞讯号传递因子的交互作用,以揭示转录调控的分子机制。我们运用多种实验技术,包括单颗粒冷冻电子显微镜 (cryo-EM)X 光晶体学、生化与生物物理方法,以及 AI 辅助的计算建模,来研究这些复合体的结构与调控机制。
